摘要
目的:观察淫羊藿总黄酮(TFE)对病毒性心肌炎小鼠免疫功能的影响。方法:雄性BALB/C小鼠32只,随机分为正常组、模型组、TFE高剂量组和TFE低剂量组,每组8只。模型组和TFE高、低剂量组采用腹腔接种柯萨奇病毒B3的方法建立病毒性心肌炎模型。末次接种1h后,正常组及模型组小鼠灌胃羧甲基纤维素钠溶液,TFE高、低剂量组分别灌胃200、100mg/kgTFE。连续灌胃14d后,测定心肌肥厚指数,HE染色法观察心肌病理变化,比色法测定血清LDH、CK水平,ELISA法检测血清IFN-γ、IL-2、IL-4、IL-10水平。结果:4组小鼠心肌肥厚指数,血清LDH、CK、IFN-γ、IL-2、IL-4及IL-10水平差异均有统计学意义(F=42.895,105.240,96.955,15.679,12.661,25.453,13.925,P<0.001)。与正常组比较,模型组小鼠心肌发生明显病理改变,心肌肥厚指数和血清LDH、CK、IFN-γ、IL-2水平上升,而IL-4、IL-10水平下降(P<0.05)。与模型组比较,2个TFE组心肌病理变化减轻,心肌肥厚指数和血清LDH、CK、IFN-γ、IL-2水平降低,IL-4、IL-10水平升高(P<0.05)。高剂量组效果较明显。结论:TFE可通过免疫调节保护病毒性心肌炎小鼠心脏。
Aim:To observe the influence of total flavones of epimedium(TFE) on immunological regulation of viral myocarditis mice.Methods:A total of 32 male BALB/C mice were randomly divided into normal,model,TFE high-dose and TFE low-dose groups,and eight in each group.The viral myocarditis model was established with intraperitoneal inoculation of coxsackie virus B3 in model,TFE high-dose and TFE low-dose groups.1 h after the last inoculation,carboxymethlylcellulose sodium solution was irrigated to the mice in model and normal groups.200 and 100 mg/kg TFE was irrigated to the mice in TFE high-and low-dose groups for 14 d.Myocardial hypertrophy index was detected,pathologic changes of myocardium were observed with HE staining,serum LDH and CK levels were assayed with colorimetric method,and serum IFN-γ,IL-2,IL-4,IL-10 levels were determined by ELISA method.Results:There were significant differences in these indices mentioned above among the four groups(F=42.895,105.240,96.955,15.679,12.661,25.453,13.925,P〈0.05).Compared with those of the normal group,the pathologic changes of myocardium happened,myocardial hypertrophy index,serum LDH,CK,IFN-γ,IL-2 levels obviously rised,while serum IL-4,IL-10 levels significantly decreased in the model group(P〈0.05).Compared with those of the model group,the pathologic changes of myocardium improved,the myocardial hypertrophy index,serum LDH,CK,IFN-γ,IL-2 levels decreased,and serum IL-4,IL-10 levels increased in the two TFE groups(P〈0.05).The effectiveness of TFE high-dose group was more significant.Conclusion:TFE could protect the hearts of viral myocarditis mice through immunological regulation.
