摘要
为探讨HL60/HHT细胞系耐药性的分子机理,采用反复短期暴露法建立HL60/HHT细胞系,并检测其bcl-2和bax的含量。结果表明,建立了多药抗性HL60/HHT细胞,其bcl-2水平为HL60的1.50倍(92.05/61.35),有显著性差异(P<0.01)。其bax蛋白在二种细胞中均低水平表达(HL6015.92,HL/60/HHT17.63),差异无显著意义(P>0.05)。因此。
To probe into the molecullar mechanism of drug resistance of HL60/HHT cell lines. the repeat and short term exposing method was used to develop the drug resistant cell lines. Using FITC Conjugated monoclonal mouse antihuman oncoprotein to determine bcl 2 and bax protein in HL60 and HL60/HHT cells. Results show, multidrug resistant HL60/HHT sublines was established. bcl 2 protein level in HL60/HHT was 1 50 times to that in HL60 (92.05/61 35), and there was a significant difference (p<0 01) Bax protein was low level expression in both two cell lines (HL60 15.92, HL60/HHT 17 63). There was no significant difference(p>0 05). Concludes that high level of bcl 2 protein in HL60/HHT cell line is one of the important causes of its nonsensitivity.
