寡聚糖激发子对杨树抗溃疡病的诱导作用 被引量:1

摘要 【目的】研究寡聚糖激发子对毛白杨愈伤组织抗杨树溃疡病菌的诱导作用。【方法】分别采用筛选出降解杨树溃疡病菌丝体、壳聚糖、果胶糖且活性较高的寡聚糖激发子A1、B、C3诱导毛白杨愈伤组织,以无菌水处理的毛白杨愈伤组织为对照,并在诱导48h后挑战接种杨树溃疡病菌,测定毛白杨愈伤组织内Vc、可溶性蛋白质和可溶性糖含量的变化。【结果】经A1、B、C3 3种寡聚糖激发子诱导48h后接种杨树溃疡病菌,毛白杨愈伤组织中Vc、可溶性蛋白质含量的变化基本上呈单峰曲线,A1、B、C3 3种寡聚糖激发子诱导接种的毛白杨愈伤组织中Vc含量峰值比对照峰值分别提高了99.02%,95.46%和85.64%;寡聚糖激发子诱导接种的毛白杨愈伤组织中可溶性蛋白质含量在发病初期(0~48h)比对照高,在发病后期(60~96h)均缓慢下降,而对照毛白杨愈伤组织中的可溶性蛋白含量在60h时上升到最高峰(4.48mg/g),均高于3种寡聚糖激发子诱导接种毛白杨愈伤组织,之后急剧下降,至96h时为2.33mg/g,比3种寡聚糖激发子诱导接种毛白杨愈伤组织都低;4个处理毛白杨愈伤组织中可溶性糖含量的变化呈双峰曲线,寡聚糖激发子A1、B、C3诱导接种的毛白杨愈伤组织中可溶性糖含量最大值分别比对照的最大值高42.4%,27.6%和16.5%。方差分析表明,A1、B、C3 3种寡聚糖激发子对挑战接种毛白杨愈伤组织中的Vc、可溶性蛋白质和可溶性糖含量均有极显著影响(P<0.01)。【结论】在一定质量浓度范围内,寡聚糖激发子可诱导毛白杨愈伤组织中的Vc、可溶性蛋白质和可溶性糖含量明显提高,从而增强了其抗杨树溃疡病菌入侵的能力。 【Objective】The study aimed to research the induction of oligosaccharides elicitors to Dothiorella gregaria resistance in Populus tomentosa callus.【Method】The Chinese white poplar callus were induced by three oligosaccharides elicitors including elicitor A1,elicitor B and elicitor C3 which had high activity and respectively were selected through the hydrolysis of D.gregaria,chitosan-oligosaccharides and pectose-oligosaccharides,with aseptic water as the CK.First the poplar callus were induced by oligosaccharides elicitors for 48 hours,then the inducted and CK callus were challengely inoculated with D.gregaria.The content variations of Vc,soluble protein,soluble sugar of poplar callus were assayed.【Result】The content changes of Vc and soluble protein in callus inoculated with D.gregaria after induced by elicitor A1,B and C3 for 48 hours all were essentially a single-peak curve.The peak contents of Vc in induced poplar callus respectively increased by 99.02 %,95.46% and 85.64% compared with that of CK.The contents of soluble protein in induced poplar callus were higher than that of CK at early pathogenesis(0-48 h) and they were all slowly decreased in induced callus at the late pathogenesis(60-96 h),meanwhile,in CK callus they reached the highest peak(4.48 mg/g) which was higher than that in three induced callus at 60 hours.Afterwards,the contents of soluble protein in CK callus dramatically decreased to 2.33 mg/g and was lower than that of all three treatment callus at 96 hours.The contents of soluble sugar in four treatment callus showed a double-peak curve.The peak contents of soluble sugar in callus inoculated with D.gregaria after induced by elicitor A1,B and C3 for 48 hours were respectively 42.4%,27.6% and 16.5% higher than that of the CK.Variable analysis indicated that the variations of content of Vc,soluble protein and soluble sugar of callus inoculated with D.gregaria after induced by three elicitors A1,B and C3 all showed highly significant differences(P0.01).【Conclusion】In certain concentration range,the contents of Vc and soluble protein were improved obviously in the poplar callus induced by oligosaccharides elicitors,so that the oligosaccharides elicitors can enhance the disease resistance of poplar celles.
