胡椒碱对人肝癌HepG2细胞抗肿瘤活性的体外实验研究 被引量:11

摘要 目的探讨胡椒碱(piperine)对人HpeG2肝癌细胞株的增殖、杀伤和细胞凋亡的影响,为肝癌的治疗提供理论依据。方法体外培养的HpeG2细胞,采用MTT比色法检测不同浓度胡椒碱对体外培养的HepG2细胞和新分离的外周血白细胞的增殖抑制作用。Hoechst 33258染色观察细胞凋亡形态,采用流式细胞术测定胡椒碱对HepG2细胞的凋亡。结果胡椒碱对HepG2细胞增殖的抑制率随着浓度的升高而增加,半量抑制浓度(IC50)为15.13±3.21μmol/L,低于其对外周血白细胞的抑制率(64.52±5.32μmol/L)。Hoechst 33258染色后,胡椒碱处理癌细胞组表现出典型的细胞凋亡特征,流式细胞仪检测20μmol/L的胡椒碱处理HepG2细胞24h后,细胞凋亡率由对照组的2.89%上升到了21.76%。结论胡椒碱具有抑制HepG2细胞增殖和诱导凋亡的抗肿瘤活性,有应用于肝癌治疗的潜在价值。 Objective To study the inhibitory effect of piperine on HepG2 cell in vitro. Methods The human hepatocarcinoma HepG2 cells were cultured in vitro and divided in, to control group and piperine-treated group. MTT assay was performed to evaluate the proliferation inhibitory effects of piperine on HepG2 cells and freshly prepared peripheral white blood cells. Hoechst 33258 nuclear staining was performed to detect the mode of cell death. Flow cytometry was used to assess the effects of piperine treatment on cell apoptosis in human HepG2 cells. Results The inhibitory effect of piperine treatment on, HepG2 cell proliferation increases with the dose, and the half inhibition concentration(IC50 ) was 64.52±5.32μmol/LL, which was lower than that for peripheral white blood cell(IC50 = 64.52 ± 5.32 μmol/L). Piperine treated HepG2 cells showed typical apoptotic characteristics. After treated with 20 μmol/L piperine for 24 h,the apoptosis rate increased from 2. 890/60 of control group to 21.76%. Conclusion Piperine has cytotoxicity effect on cultured human HepG2 cells in vitro,and can inhibit proliferation and induce apoptosis.
