金银花提取物体外抗猪繁殖与呼吸综合征病毒研究 被引量:21

摘要 通过观察金银花提取物对猪繁殖与呼吸综合征病毒(PRRSV)感染的Marc-145细胞的保护效果、对病毒感染滴度(TCID50)的影响及对ORF7mRNA表达的影响来评价其体外抗猪繁殖与呼吸综合征病毒的活性。结果表明,金银花提取物大孔树脂600mL/L的乙醇/水洗脱部位对PRRSV感染细胞具有较好的保护效果,最小保护浓度为6.25μg/mL。6.25μg/mL 600mL/L的乙醇/水洗脱部位使PRRSV病毒滴度从105.8 TCID50降低至100.3 TCID50左右,使PRRSV ORF7mRNA含量降低1 000多倍。因此,金银花提取物大孔树脂600mL/L的乙醇/水洗脱部位具有良好的体外抗PRRSV活性。 The antiviral effect of Lonicera japonica on PRRSV in vitro was evaluated by observing cytopathic effect(CPE) on PRRSV infected Marc-145 cell,determining TCID50 and evaluating ORF7 mRNA expression level.The results showed that the 600 mL/L ethanol/water elution fractions possess the perfect protective effect on PRRSV infected cells and with the minimum protective concentration of 6.25 μg/mL.The 6.25 μg/mL 600 mL/L ethanol/water elution fractions decreased PRRSV titer from 105.8 TCID50 to 100.3 TCID50 and reduced the PRRSV ORF7 mRNA contents about 1 000 times.In summary,we disclosed 600 mL/L ethanol/water elution fractions of Lonicera japonica as a potential antiviral drug owing to its perfect anti-PRRSV effect.
