摘要
目的 探讨腺病毒介入性血管内皮生长因子 (VEGF)基因导入皮下促进血管新生的可行性。方法 使用2 5G 1ml注射器将 0 .2ml含有 1× 10 9pfu( plaqueformationunit)腺病毒AdexCAhVEGF165注入小鼠腹部正中皮下。对照组注入对照腺病毒AdexCAlacZ于小鼠皮下 ,15天后处死动物 ,组织学切片随机观察皮下组织中毛细血管数量。结果 观察组皮下组织可见大量新生血管。每平方毫米含 2 2 0± 44个毛细血管 ,较对照组每平方毫米 70± 14个增加约 3倍 (t =8.4,P <0 .0 1)。结论 VEGF导入皮下可增加皮下组织毛细血管数量 ,可望用于缺血性疾病的治疗。
Objective To explore the feasibility of increasing neovascularization by adenovirus mediated transfer a vascular endothelial growth factor(VEGF) gene into rat subcutaneous tissue.Methods 0.2 ml of AdexCAVEGF 165 containing 1×10 9 pfu adenovirus was injected into subcutaneous tissue of rat abdoman.Tissue section was processed to observe the number of capillary at 15th day after gene transfer.Results 220±44 capillary/mm 2 was randomly observed in subcutaneous tissue of rat subjected to injection of AdexCAVEGF 165 ,much higher than that(70±14 capillary/mm 2) of rat subjected to injection of control adenovirus (P<0.01).Conclusion Our data showed that VEGF could enhance neovascularization in rat subcutaneous tissue,suggesting VEGF might be used to treat ischemic diseases in the future.
