端粒酶活性及其hTR表达在诱导大肠癌分化细胞中的变化 被引量:4

摘要 【目的】观察全反式维甲酸 (ATRA)和 1,2 5 二羟维生素D3 (VD3 )大肠癌细胞株诱导分化过程中端粒酶活性的变化 ,并对其RNA组分hTR进行检测。【方法】ATRA和VD3 对大肠癌细胞进行诱导分化 ,在进行细胞活力测定、细胞增殖测定、流式细胞仪 (FCM )分析细胞周期及碱性磷酸酶比活性测定的同时 ,分别在诱导后第 2天、第 4天和第 6天用TRAP法检测其端粒酶活性的变化 ,并同时用RT PCR的方法对端粒酶的RNA组分进行检测 ,PAGE观察结果。【结果】TRAP法检测端粒酶活性 ,结果显示在诱导分化的第 2天后活性减弱 ,第 4天、第 6天其活性受到明显抑制。利用RT PCR法检测各阶段端粒酶RNA组分无明显改变。【结论】ATRA和VD3 诱导大肠癌细胞分化过程中端粒酶活性明显抑制 ,但RT PCR检测其RNA组分无改变。 Objective To observe the change of telomerase activity and hTR expression in differentiating colon cancer cells induced by ATRA and VD 3. Methods The colon cancer cells were induced and differentiated by ATRA and VD 3 and their telomerase activity and RNA expression were tested on the secondary, fourth and sixth day respectively. PAGE was used to observe the results. Result The telomerase activity is inhibited gradually on secondary, fourth and sixth day, but RNA expression didn't change. Conclusion The telomerase activity is inhibited obviously in differentiating colon cancer cells induced by ATRA, VD 3, but their RNA components keep constant using RT PCR for detection.
