摘要
【目的】观察血管紧张素Ⅱ对心肌细胞内皮型一氧化氮合酶 (eNOS)表达的影响。【方法】用逆转录聚合酶链式反应 (RT -PCR)检测新生大鼠心肌细胞eNOSmRNA(以GAPDH为内标 )水平。【结果】在培养新生大鼠心肌细胞和非心肌细胞中均能检测到eNOSmRNA ,其中心肌细胞的表达高于非心肌细胞 ;血管紧张素Ⅱ作用 6、12和 2 4h ,心肌细胞eNOSmRNA水平明显减少。【结论】在大鼠心肌细胞和非心肌细胞皆有eNOS基因表达 ;血管紧张素Ⅱ可抑制心肌细胞eNOS基因的表达。
Objective To observe the effect of angiotensin Ⅱ on the expression of endothelial type nitric oxide synthase gene in cultured cardiac myocytes. Method The endothelial type nitric oxide synthase (eNOS) and glyceraldehyde 3 phosphate dehydrogenase (GAPDH) specific primers were designed according to their cDNA sequences. The eNOS gene expression was assessed using reverse transcription polymerase chain reaction (RT PCR) standardized with GAPDH. Result The eNOS mRNA in cultured neonatal rat cardiac myocytes and non myocytes was detected; the eNOS mRNA levels in cardiac myocytes were higher than in non myocytes; treatment with angiotensin Ⅱ for 6, 12 and 24 h decreased signifcantly the eNOS mRNA levels in cardiac myocytes. Conclusion eNOS gene is expressed in cultured neonatal rat cardiac myocytes and nonmyocytes. Angiotensin Ⅱ can inhibit eNOS gene expression in a time dependent manner in cultured cardiomyocytes.
