摘要 【目的】建立人增生性瘢痕 (HTS)裸鼠模型和观察瘢痕组织形态学变化特点。【方法】将HTS移植至裸鼠的皮下 ,采用光镜、电镜和免疫组化方法观察移植后的HTS的形态学、T淋巴细胞、巨噬细胞和转化生长因子 β1(TGF β1)的分布变化特点。【结果】术后裸鼠全部成活 ,植入的瘢痕无坏死。在 6~ 7周内均能保持其原有的组织结构。光镜、电镜观察成纤维细胞 (FB)内含丰富的粗面内质网、线粒体 ;移植后瘢痕组织中的T淋巴细胞、巨噬细胞细胞分布和TGF β1免疫反应强度与移植前相似。【结论】在裸鼠体内建立的HTS模型是一个可行和可靠的动物模型 ,移植的HTS在 6~ 7周内仍能保持其原有的组织形态特征。 Objective To establish a model of human hypertrophic scars in nude mice and to investigate its morphological characteristics of the scars. Methods Human hypertrophic scars were implanted subcutaneously in the nude mice. The morphological changs were observed using light and electron microscopes. T lymphocytes,macrophages and TGF β 1 in the implanted scars were demonstrated by immunohistochemical staining ,and the variation were analyzed Results All the mice survived after implantation of human hypertrophic scars. No necrosis was found in the implanted scars under the light microscopy.The original morphology of the scars was maintained at sixth and seventh weeks after transplantation .Demonstrated by electron microscopy,the fibroblasts in the scars contained rich rough endoplasmic reticula and mitochondria.There were no changes of density of infiltrated T lymphocytes and macrophages. Immunohistochemically,TGF β 1 reaction was stronger in the scars implanted than in control. Conclusions Implantation of human hypertrophic scars in nude mice was feasible and reliable animal model for scar study.There is no morphological variation in the human hypertrophic scars in nude mice until 6~7 weeks after implanting.
