摘要
目的研究瑞芬太尼对大鼠脑缺血再灌注损伤后bcl-2和Caspase-3表达的影响,探讨瑞芬太尼对脑缺血再灌注损伤的作用。方法以线栓法制作大鼠大脑中动脉阻塞的脑缺血再灌注模型,采用免疫组化、Real-time PCR法和神经行为检测相结合的方法,观察缺血再灌注侧大脑皮质内bcl-2和Caspase-3的表达及神经功能的变化。结果与缺血再灌注组比较,瑞芬太尼组的bcl-2mRNA实时表达明显增高(P<0.05),Caspase-3mRNA实时表达明显降低(P<0.05),神经异常行为明显减轻。结论瑞芬太尼可上调大鼠脑缺血再灌注损伤后bcl-2的表达,下调Caspase-3表达,明显改善神经症状,对脑缺血再灌注损伤有神经保护作用。
Objective To evaluate the effect of Remifentanil on the expression of bcl-2 and Caspase-3 in rats cerebral cortex following ischemia-reperfusion injury.Methods The cerebral ischemia-reperfusion model was established by thread embolism of the middle cerebral artery.Immunohistochemistry,real-time PCR and neurological evaluation were used to examine the expression of bcl-2 and Caspase-3 in cerebral cortex and neurological function disorders.Results As compared with ischemia-reperfusion group,the relative mRNA level of bcl-2 was increased significantly(P0.05),that of Caspase-3 was decreased significantly(P0.05),and neurological function disorders were markedly relieved in Remifentanil group.Conclusion Remifentanil has a neuropretective effect on ischemia-reperfusion injury by increasing the expression of bcl-2,decreasing the expression of Caspase-3 and improving neurological symptoms.
