摘要
采用高速逆流色谱和大孔吸附树脂相结合的技术,对冻青中的黄酮类化合物进行分离纯化。高速逆流色谱溶剂体系为正己烷-乙酸乙酯-甲醇-水(3∶4∶5∶3,体积比),上相为固定相,下相为流动相,螺旋管柱正转800 r.min-1,流动相流速3 mL.min-1,进样量20 mL,进样浓度20 mg.mL-1,检测波长254 nm。分离获得两种高纯度黄酮类化合物,纯度分别为98.01%、96.10%,经HPLC和NMR鉴定为鼠李秦素-3-O-β-D-葡萄糖苷和高圣草素-7-O-β-D-葡萄糖苷。
High-speed counter-current chromatography (HSCCC) combined with macroporous adsorption resins was employed to separate and purify flavones from Viscum coloratura (Kom.)Nakai. The two phase sol- vent system of HSCCC consisted of n-hexane-ethyl acetate-methanol-water(3 : 4 : 5 : 3,volume ratio) with the stationary phase as its upper layer and the mobile phase as its lower layer,coiled column rotate speed was 800 r ~ min-1 ,flow rate was 3 mL . min-1 ,injection volume was 20 mL,sample concentration was 20 mg. mL-1 ,detection wavelength was 254 nm. Two high-purity compounds, rhamnazin-3-O-β-D-glucoside and homoeriodictyol- 7-O-β-D-glucoside were obtained with purity of 98.01%, 96.10 %, respectively. And their structures were identified by HPLC and NMR.
