摘要
目的:优选黄花草木犀总皂苷最佳提取纯化工艺。方法:以黄花草木犀总皂苷含量为考察指标,采用正交试验,研究黄花草木犀最佳提取和纯化工艺。结果:最佳提取工艺为加14倍量70%乙醇提取3次,每次1 h;最佳纯化工艺为浓缩浸膏上D101型大孔树脂柱,洗脱剂70%乙醇,吸附流速1 mL.min-1,洗脱流速3 mL.min-1,洗脱体积6 BV。结论:黄花草木犀总皂苷优选的提取纯化工艺简便、稳定、可靠。
Objective:To optimize optimum extraction process and purification process of total saponins from Melilotus officinalis.Method:Orthogonal test was designed for investigating optimum extraction and purification technology for M.officinalis with the content of total saponins as index,respectively.Result:Optimum extraction process of total saponins were:extracted 3 times with 14 times the amount of 70% ethanol for 1 h each time,extraction solvent to reflux for 1.0 hour and 3 times.Optimum purification process was:concentrated extract passed through D101 resin with eluting by 6.0 BV 70% ethanol,elution flow rate was 3.0 mL·min-1,adsorption flow rate was 1.0 mL·min-1.Conclusion:This optimized process was simple,stable and reliable.
