摘要
目的建立知母甾体总皂苷的HPLC指纹图谱,用以评价和监控提取工艺的稳定性。方法采用Agilent HC-C18(5μm,4.6 mm×250 mm)色谱柱,ELSD检测器,流动相:甲醇-水梯度洗脱,流速:1 ml.min-1,漂移管温度40℃,载气压力3.5 bar。采用直观分析和相似度软件评价知母甾体总皂苷指纹图谱的相似度。结果建立了知母甾体总皂苷的HPLC-ELSD指纹图谱,检测了10个批次的知母甾体总皂苷,标定了15个共有峰,各色谱峰分离度较好,相似度较高,达到了中药指纹图谱研究的技术要求。结论该法准确可靠,建立的HPLC-ELSD指纹图谱可为知母甾体总皂苷的质量控制提供科学准确的依据。
Objective To establish HPLC-ELSD fingerprint of anemarrhenasaponin. Methods The separation of anemarrhenasaponin was performed on a Agilent HC-C18(5 μm,4.6 mm×250 mm) column of HPLC.The mobile phase was methanol-water and the flow rate was 1 ml·min-1.The detector was ELSD.The temperature of drift tube was 40℃ and the press was 3.5bar.The data were analyzed by Fingerprint Similarity Evaluation Software to compare the similarity of the samples from different batches. Results Fingerprint spectrum of anemarrhenasaponin was established.10 samples were detected and 15 peaks in the chromatogram were common.There was a high similarity and each chromatographic peak was obtained with good separation and correlation according to the technical requirements of fingerprint of Chinese traditional medicine. Conclusion This method shows high precision and reliability and the fingerprint spectrum provides a scientific basis for controlling the quality of anemarrhenasaponin.
