摘要
目的研究桑叶提取物对α-葡萄糖苷酶活性的影响。方法采用葡萄糖氧化酶法,利用蔗糖为底物,以酵母产α-葡萄糖苷酶和大鼠小肠上皮细胞(IEC-6)中酶来研究桑叶提取物对酶活性的影响。结果在酶-抑制剂模型上,不同浓度桑叶提取物(5,2.5,1.25,0.625 mg/ml)对α-葡萄糖苷酶抑制活性分别为54.62%,39.79%,8.51%,6.62%;5 mg/ml阳性药物(阿卡波糖)的抑制作用为65.60%;在IEC-6细胞模型上,不同浓度桑叶提取物(200,100,50μg/ml)对α-葡萄糖苷酶的抑制活性分别为26.88%,12.76%,5.92%;250μg/ml的阳性对照(阿卡波糖)抑制率为47.46%。结论桑叶提取物对α-葡萄糖苷酶活性具有抑制作用,随抑制率浓度增加而增加,对桑叶提取物降糖药物开发具有指导意义。
Objective To study the inhibitory effect of mulberry leaf extracts on alpha-glucosidase activity. Methods The inhibitory effects of mulberry leaf extracts on alpha-glucosidase derived from bakers yeast and small intestinal epithelial cells(IEC-6)were measured by using oxidase colorimetric method and using sucrose as the substrate. Results In the enzyme-inhibitor model,the inhibition rates of 5,2.5,1.25,0.625mg/ml mulberry leaf extracts were 54.62%,39.79%,8.51%,6.62%,respectively.The inhibition rate of 5mg/ml positive drug(acarbose)was 65.60%.On the IEC-6 cell model,the inhibition of 200,100,50μg/ml mulberry leaf extracts were 26.88%,12.76%,5.92%,while the positive drug was 47.46%. Conclusion The results show that mulberry leaf extracts have a certain degree of α-glucosidase inhibitory activity and the inhibition increases with the increasing concentration.The results mean a lot to develop the diabetes drug.
