摘要
目的探讨荞麦黄酮复方制剂(TFBCP)对2型糖尿病大鼠心肌病的保护作用及其可能机制。方法将SD大鼠70只,随机取出12只作为正常对照组(Contorl group),其余建立大鼠2型糖尿病模型。将成模大鼠随机分为4组,即TFBCP低、高剂量组(L、H-TFBCP group)、消渴丸组(XKW group)和模型对照组(Model group)。观察空腹血糖(FBG),心重指数(HWI:hw/bw)及心功能。采用放射免疫分析法和Western-Blot法结合计算机图像分析技术,检测心肌P38α-丝裂原活化蛋白激酶(P38α-MAPK)蛋白表达的变化。并光镜下观察心室肌组织病理改变。结果与模型对照组相比,TFBCP能剂量依赖性降低2型糖尿病大鼠FBG和HWI,同时改善心功能;对P38α-MAPK蛋白表达含量的增加,及对其激活后由胞浆至胞核的活性位移,TFBCP均具有一定的抑制作用;能显著改善心肌组织结构的异常变化,其中TFBCP高剂量组与阳性药物消渴丸效果相近。结论 TFBCP对2型糖尿病大鼠心肌病具有逆转保护作用,其机制可能是拮抗了P38α-MAPK信号转导通路。
Objective To study the protection of the flavonoid of buckwheat Compound Preparation(TFBCP) against experimental diabetic cardiomyopathy in type 2 diabetes rats and its mechanism. Methods The SD rats were 70,12 were randomly selected in control group.The rests were built the Type 2 diabetes model.And then the models were randomly divided into 4 groups-L,H-TFBCP group,XKW group and model group.At the end of the experiment,the fasting blood glucose(FBG),the heart weight index(HWI:hw/bw) and the cardiac function were observed.Radioimmunoassay and Wetern-Blot techniques were used to measure P38α-mitogen activated protein kinases(P38α-MAPK) expression.Ventricular myocardium pathological changes were observed under light microscope. Results Compared with model group,TFBCP resulted in a dose-dependent decrease in FBG and HWI,improved cardiac function.TFBCP intervention group could inhibit the P38α-MAPK expression and activity displacement.Myocardium cells and myocardium's stroma changes in different dose of TFBCP groups were also significantly improved.The protection of H-TFBCP group was more effective,the role and XKW group were similar. Conclusion TFBCP has some protection on diabetic cardiomyopathy in type 2 diabetes rats.The mechanism may be related to inhibition of P38α-MAPK signal transduction system.
