丹参加玻璃酸钠预防性关节内注射对兔膝关节软骨退行性变的影响

Effects of Preventive Intra-Articular Injection of Radix Salvias Miltionrrfh1zae and Sodium Hyaluronate on Cartilage Degeneration of Osteoarthritis in Rabbit Knee Jionts

目的通过观察丹参加玻璃酸钠预防性关节内注射对兔膝关节软骨退行性变的影响,探讨其治疗作用机理。方法将80只日本大耳白兔随机分为A组、B组、C组、D组,每组20只。通过切断膝关节前交叉韧带和内侧副韧带造成膝OA模型,每组每只动物分别注入0.3 ml的生理盐水(A组)、玻璃酸钠注射液(B组)、丹参注射液(C组)、丹参加玻璃酸钠混合液(D组),手术当天给药,每周1次。于给药4、8周后分批处死实验兔,每批每组各10只。行关节腔大体观察、光镜下组织学观察、凋亡软骨细胞检测。结果各组术后4、8周组间比较,软骨退变计分、软骨凋亡细胞计数,有无显著性意义;D组与B、C组之间差异无显著性,但D组的均值小于B、C组(P>0.05);各组术后4、8周组内比较,B、C、D组软骨退变计分、软骨凋亡细胞计数相比较,差异无显著性(P>0.05);A组软骨退变计分、软骨凋亡细胞计数相比较,有显著性差异(P<0.05)。结论在OA早期,预防性关节腔注射丹参和HA,能明显延缓关节软骨的蜕变,为早期防治OA提供实验基础。

Objective To observe the preventive effects of intra-articular injection of Radix Salviae Miltiorrhizae and Sodium Hyaluronate against cartilage degeneration of osteoarthtitis rabbit knee joint and the mechanism of knee osteoarthritis. Methods Knee osteoarthritis models were created by cuting off the anterior cruciate ligament and medial collateral ligament in the knee joint,80 adult Japanese white rabbits were divided randomly into 4 groups： Group A,Group B,Group C,Group D.After taking drug for 4 and 8 weeks,the rabbits were killed and the joints gross observing,histological analysis and examination of cell apoptosis were done. Results After 4 and 8weeks,the rabbits were killed,and the inferior articular cartilage of the femoral condoyle was got for histological analysis and examination of cell apoptosis.The score of articular cartilage degeneration and the number of apoptosis cells of cartilage in group B,C or D were smaller than that in group A,and the mean score in group D was smaller than that in group B or C（P0.05）.With the same B,C or D group was compared,the score of articular cartilage degeneration and the number of apoptosis cells of cartilage after 4 and 8 weeks had no significant difference（P0.05）,and with the same A group was compared the score of articular cartilage degeneration and the number of apoptosis cells of cartilage after 4 and 8 weeks had significant difference（P0.05）. Conclusion In early OA,preventive intra-articular injection of Radix Salviae Miltiorrhizae and Sodium Hyaluronate can significantly delay cartilage degeneration,and provide an experimental basis for early prevention and treatment of OA.
