摘要
目的研究乌贼墨肽聚糖的提取和分离纯化工艺,探讨其基本的结构组成,并研究它的抗前列腺癌活性。方法用Tris-HCl缓冲液提取、丙酮脱脂和Sevag试剂除蛋白方法提取出粗肽聚糖。将提取物用超滤、DEAE-52离子交换柱、G-75凝胶柱进行分离纯化,高效液相色谱和SDS-PAGE凝胶电泳进行纯度检测,经红外扫描,确定该组分具有肽聚糖的一般吸收特征,以及检测它的氨基酸组成。最后采用MTT法检测纯化后得到的肽聚糖对DU-145和PC-3细胞的增殖的影响。结果最终水提得到的肽聚糖的纯化物对DU-145和PC-3细胞均有较明显的增殖抑制效应,且抑制作用具有明显量效依赖关系。结论该法提取的乌贼墨肽聚糖具有明显的抗前列腺癌作用。
Objective To extract the anti-prostatic cancer(Pca) effect of peptidoglycan from ink of Sepia.. Methods The peptidoglycan was extracted by Tris-HCl buffer solution,acetone degreasing and sevag reagent except protein.Peptidoglycan was purified by ultrafiltration,DEAE-52 and Sephadex G-75 column chromatography and the purity of peptidoglycan was demonstrated by HPLC and SDS-PAGE electrophoresis,and structure characterization by IR.The anti-Pca peptidoglycan were evaluated by MTT. Results The results of MTT showed that peptidoglycan from ink of Sepia significantly inhibited the proliferation of DU-145 and PC-3 cells. Conclusion The peptidogly can from the ink of Sepia shows obvious anti-Pca activity.
