摘要
目的:了解卵巢切除(OVX)大鼠胫骨对氟化钠和尼尔雌醇联合应用的治疗反应。方法:将24只雌性大鼠随机分成3组:OVX组、对照组和EF组,每组8只。对OVX组和EF组大鼠行OVX手术,建立骨质疏松动物模型。EF组于OVX术后1个月予氟化钠和尼尔雌醇联合治疗,给药3个月后处死。用骨形态计量学方法检测OVX大鼠左后肢胫骨对氟化钠的治疗反应。结果OVX组大鼠骨小梁体积、平均骨小梁密度较对照组显著减少(P<0.05);四环素标记表面、类骨质表面和组织水平的骨形成速率均较对照组显著增加(P<0.05);矿化沉积速率、类骨质成熟时间和矿化延迟时间,较对照组缩短(P<0.05)。EF组治疗12周后发现,骨小梁体积和平均骨小梁宽度高于OVX组(P<0.05),其中TBW显著高于对照组(P<0.05);骨骺部位MTPD和MTPS与对照组比较无显著差异(P>0.05)。EF治疗组骨小梁的四环素标记表面、类骨质表面、组织水平骨形成速率和平均类骨质宽度低于OVX组(P<0.05),矿化沉积速率和矿化延迟时间低于对照组(P<0.05)。结论:尼尔雌醇和氟化钠联合治疗可显著增加平均骨小梁宽度和骨小梁骨量,并改善小梁骨空间结构。骨骺部位的改善作用较为明显。
Objective: To investigate the response of trabecular bone in ovariectomized(OVX) rats to sodium fluoride and nylestri-ol. Methods: Twenty-four female Wistar rats were randomly divided into three groups, 8 rats in each: sodium fluoride and nylestriol treated group(EF) ,OVX and control(C)group. After being fed with sodium fluoride and nylestriol for 3 months, all rats were sacrificed. The effects of sodium fluoride and nylestriol on bone microarchitecture and dynamics were studied by bone histomorphometry. Results: The data suggested that trabecular bone volume,mean trabecular width,density and cortical thickness were remarkably reduced in OVX group (P < 0.05) while tetracycline labeled surfaces, osteoid surfaces and bone formation rate were increased in comparison with those in C group. In EF group,however,the trabecular bone volume and mean trabecular width were much more increased than those in OVX group, while mean trabecular width was increased in comparison with that in C group, tetracycline labeled surfaces,osteoid surfaces,mean osteoid seam width and bone forma- tion rate in OVX group were decreased in comparison with those in C group,while mineral apposition rate and osteoid min-eralization lag time were decreased in comparison with those in C group (P < 0.05). Conclusions: Trabecular bone volume, mean trabecular width and spatial architecture were improved by the administration of sodium fluoride and nylestriol in OVX rats. The effects on epiphysis seem to be more significant than those on metaphysis.
