摘要
目的探讨高迁移率族蛋白A1(high mobility group protein A1,HMGAl)在人类甲状腺乳头状癌组织中的表达及临床意义。方法应用RT—PCR检测32例甲状腺乳头状癌组织及相应的癌旁正常组织和15例甲状腺腺瘤组织中HMGAl mRNA的表达情况;应用免疫组织化学法检测石蜡包埋的60例甲状腺乳头状癌组织和20例甲状腺腺瘤组织中HMGAl的蛋白表达水平。结果HMGAl mRNA在甲状腺乳头状癌组织中的表达量显著高于癌旁正常甲状腺组织(P〈0.01)及甲状腺腺瘤组织(P〈0.05)。HMGAl蛋白在甲状腺乳头状癌组织中的阳性表达率显著高于甲状腺腺瘤组织(P〈0.01),在有淋巴结转移组的阳性表达率高于无淋巴结转移组(P〈0.05);HMGAl蛋白的表达与患者的性别、年龄、肿瘤大小、包膜侵犯及TNM分期均无关(P〉0.05)。结论HMGAl的异常表达可能与甲状腺乳头状癌的发生发展和淋巴结转移有关,有望成为甲状腺乳头状癌诊治中的一个新的标志物。
Objective To investigate the expression and clinical significance of high mobility group protein A1 ( HMGA1 ) in human papillary thyroid carcinoma. Methods The expression of HMGA1 mRNA was detected by Reverse Transcription Polymerase Chain Reaction (RT-PCR) in tissues of 32 cases of papillary thyroid carcinoma, the corresponding adjacent normal tissues, and in 15 cases of benign thyroid lesion tissues. The expression of HMGA1 protein was detected by immunohistochemical method in 60 cases of papillary thyroid carcinoma tissues and 20 cases of thyroid adenoma tissues, with all the tissues paraffin-embedded. Results The expression of HMGA1 mRNA was significantly higher in papillary thyroid carcinoma than in the adjacent normal tissues ( P 〈 0.01 ) and thyroid adenoma tissues ( P 〈 0.05 ). The positive expression rate of HMGA1 protein was significantly higher in papillary thyroid carcinoma than in thyroid adenoma tissues( P 〈 0.01 ). The positive expression rate of HMGA1 protein was higher in the group with lymph node metastasis than in the group without lymph node metastasis (P 〈 0.05 ). The expression of HMGA1 protein in papillary thyroid carcinoma was not related to patients' sex, age, tumor size, capsule invasion or TNM stage ( P 〉 0.05 ). Conclusion The abnormal expression of HMGA1 may be associated with the oncogenesis and progress of papillary thyroid carcinoma and lymph node metastasis, which is expected to become a new marker for diagnosis and treatment of papillary thyroid carcinoma.
