摘要
目的研究抗人去整合素金属蛋白酶15(adisintegrinandmetalloproteinase15,ADAMl5)多克隆抗体(anti.ADAMl5抗体)与胃癌、肺癌和血管内皮细胞结合的特性。方法制备并纯化anti.ADAMl5抗体,用间接ELISA法检测不同时间、不同抗体滴度的情况下,肺癌A549细胞、胃癌HGC-27细胞及血管内皮HMEC.1细胞与抗体结合后的光密度值。结果抗体浓度为2-8pg/mL的条件下,anti-ADAMl5抗体与三种细胞株的结合程度随抗体浓度的增高而增高;孵育时间为24h或48h的结合程度明显高于2h。结论anti-ADAMl5抗体能与胃癌细胞、肺癌细胞和血管内皮细胞结合,有望用于恶性肿瘤的诊治。
Objective To study the combinations of anti-human ADAM15 polyclonal antibody with gastric cancer, lung cancer, and vascular endothelial cell.Methods Anti-ADAM15 antibody was purified and then we detected the combinations of this antibody with lung cancer cell A549,gastric cancer cell HGC-27,and vascular endothelial cell HMEC-1 under different time or different concentrations using an indirect ELISA method.Results When the final concentrations of this antibody were from 2 to 8 I~ g/mL,Anti-ADAM15 antibody effectively combined with both tumor cell and vascular endothelial cell;the combination became stronger when the time of reaction was long enough(24 or 48h) or the concentration of this antibody became higher. Conclusion Anti- ADAM15 antibody can combine with gastric cancer cell,lung cancer cell and vascular endothelial cell,and may be used in diagnosis or treatment of malignant tumor.
