冷诱导下毛百合甘油-3-磷酸酰基转移酶活性变化及其基因保守区的克隆 被引量:8

摘要 探讨了野生毛百合甘油-3-磷酸酰基转移酶活性在冷胁迫下的变化,用简并引物扩增了甘油-3-磷酸酰基转移酶基因保守区序列的结果表明,该保守区段长744 bp,推测编码247个氨基酸。在GPAT氨基酸序列中存在1个高度保守的区域(WIAPSGGRDRP),在NCBI上经过BLASTp比对分析,发现这段保守区序列为LPLAT基因超级家族酶类的催化活性区,此家族多为催化酰基辅酶A(acylCoAs)或者酰基载体蛋白(acylACPs)中的酰基与受体蛋白结合的酰基转移酶类。此序列Genbank登录号为HQ654523。 Change in Glycerol-3-Phosphate Acyltransferase (GPAT)activity of Lilium dauricum under cold stress was explored and the conserved sequence of GPAT was amplified using degenerate primers. The results demonstrated that the conserved sequence was 744 bp in length,which was deduced coding 247 amino acids. There is a highly conserved domain (WIAPSGGRDRP)in the amino acid sequence of GPAT. Aligning the conserved domain with BLAST of NCBI,we found it was a catalytic domain of LPLAT gene superfamily enzymes. This family mostly catalyzed acyl-coenzyme A (acylCoAs)or acyhransferase which was acyl in the Acyl carrier protein(acylACPs) binding with receptor protein. The gene accession nucleotide sequence number in GenBank was bankit1413285.
