摘要
克隆了与草莓成熟有关的乙烯受体Ers1基因片段,为进一步研究Ers1基因功能并通过基因技术改善草莓贮运性能奠定基础。以全明星草莓成熟果实中分离得到基因组DNA为模板,经PCR扩增到一条约700 bp的特异片段,将该片段克隆到PEGM-T easy vector上经测序分析,基因全长667 bp,编号222个氨基酸。序列分析结果表明,该序列与Chandler-Ers1的cDNA序列同源性99%,氨基酸序列同源性98%。
In this study a ripening related ethvlene recepor Ersl gene in strawberry fruits was cloned for the following-up reseach on gene and improving varieties of strawberry through transgenic technology. A pair of primers were designed and synthesized based on the sequence reported of Chandler-Ersl. Using genome DNA extracted from All-star strawberry mature fruits as template the specific PCR product of Ersl was obtained, then it was ligated to pEGM-T easy vector and sequenced. The sequencing data showed that the PCR product was 667 bp encoding 222 peedicted amino acid residues. Comparison with the eDNA sequence from Chandler-Ersl indicated the homlogy was 98% ,and amino acid identitise were 97%.
