摘要
旨为制备兔抗类猪圆环病毒因子P1的特异性抗体,对其在病原学方面的应用进行研究。采用人工合成选定的表位肽,将其与载体蛋白KLH偶联后免疫新西兰大白兔制备抗P1多克隆抗体,通过免疫组化对该多抗与P1的反应性进行检测。获得了抗P1多克隆抗体;免疫组化结果显示抗体可与P1病毒蛋白出现特异性反应。偶联后的P1表位肽具有免疫原性,可用于制备相应的抗体;制备的抗体特异性良好,可用于P1病原学方面的研究。
To generate and investigate rabbit polyclonal antibody against porcine circovirus-like agent P1. The certain epitope peptides were chosen and synthesized, and then coupled to keyhole limpet hemocyanin(KLH) to increase antigenicity. New Zealand White Rabbits were immunized with KLH conjugated peptides. The polyclonal antibodies elicited were eharactered by ELISA and tested by immunohistochemieal techniques in etiological diagnosis of P1. The polyclonal antibodies against porcine circovirus-like agent P1 were generated successfully. Immunohistochemistry showed the antibodies could specifically react with P1 proteins. The synthesized aim peptides can be used to immunized rabbit in order to generate polyclonal antibodies against porcine circovirus-like agent,P1. The antibodies prepared can be used as potential tools to study etiological diagnosis of P1.
