摘要
目的 为研究 p5 3对乳腺癌细胞增殖和凋亡的影响 ,用重组人野生型 p5 3全长 c DNA的逆转录病毒载体转染Bcap37细胞 ,观察其增殖的凋亡变化。方法 应用 Northernblotting和 Western blotting检测 Bcap37细胞 p5 3表达变化 ,流式细胞仪计数检测细胞增殖及周期变化 ,MTT及苔盼兰计数观察紫外线刺激后细胞存活率差异 ,DNA末端原位标记技术检测细胞凋亡变化。结果 外源野生型 p5 3在Bcap37细胞表达后 ,使细胞生长速度变慢 ,克隆形成所需时间增加 ,并使细胞周期发生变化 ,主要表现在 G1 期生长抑制 ,即细胞停滞于 G1 期 ,紫外线刺激后 ,细胞存活率比对照细胞下降 2 0 %~ 40 % ;细胞凋亡明显增加 (所有数据均 P<0 .0 1)。结论 转染野生型 p5 3的 Bcap37细胞出现生长抑制 ,并加速 U
Objective To study the effect of p53 to growth and apoptosis of breast cancer cell line. Methods Bcap37 cells were transfected by retrovirus vector containing wtp53. By using Northern blotting and Western blotting, the change of p53 expression of Bcap37-p53 was detected; FCM analyzed the cell growth. Difference of cell survival was observed by MTT and trypanblau tests after UV stimulation. By using TUNEL the apoptosis of cells was tested. Results The expression of exogenous p53 inhibited the cell growth, the percentage of G1 cells was increased. After UV stimulation, the cell survival rate reduced 29 %~40 %; and apoptosis between Bcap37 neo and Bcap37 p53 were significantly different.Conclusion Bcap37 cells transfected by wtp53 showed growth inhibition and accelerated apoptosis after UV stimulation
