摘要
目的 探讨内皮素 (Endothelin ET- 1)在大肠腺癌血管形成中的作用。方法 应用免疫组织化学 SABC法 ,对 41例大肠腺癌和 2 0例腺瘤组织中 ET- 1,VEGF和 F RAg分别进行免疫特异性染色 ,并计数肿瘤组织内微血管密度 (MVD)。结果 ET- 1染色主要分布于肿瘤细胞胞浆 ,阳性肿瘤基质中血管内皮细胞及成纤维细胞也有较强的着色。腺癌中 ET- 1阳性表达率为 73.4% ,显著高于腺瘤的 45% (P<0 .0 5 )。等级相关分析表明 ,ET- 1与 VEGF及 MVD密切相关。 (rs’=0 .6 7,rs'=0 .5 9,P<0 .0 1)。值得注意的是 ,在 ET- 1和 VEGF均有表达的 2 6例大肠腺癌中 ,连续切片显示 VEGF表达阳性的区域 ,ET- 1也倾向于阳性 ,并且两者在血管内皮细胞均有表达。。结论 ET- 1参与大肠腺癌肿瘤血管形成 ,并且与
Objective To study the effect of endothelin 1(ET 1)on the angiogenesis of colorectal carcinoma.Methods Immunohistochemical analysis was carried out on archival specimens of 41 human colorectal adenocarcinoma and 20 adenomas by using antibodies against ET 1、VEGF and F Ⅷ RAg.Vessels were quantitated by light microscopy (200×),the intensity of staining for ET 1 and VEGF was assessed on a scale of 0~3+.Results ET 1 was stained mainly in the cytoplasm of tumour cells and weakly in vascular endothelial cells and fibroblasts.Expression of ET 1 correlated directly with expression of VEGF rs'=0 67( P <0 01). In all tumours that expressed both factors,serial sections showed that regions of ET 1 immunopositivity also tended to be positive for VEGF.Moreover,expression of ET 1 strongly correlated with microvessel density rs'=0 59( P <0 01).Conclusion ET 1 may act in synergism with VEGF in the growth and angiogenesis of human colorectal cancer.
