等电聚焦强度对木薯叶片蛋白质双向电泳分离结果的影响 被引量:8

Effect of Isoelectrofocusing Intensity on Protein Separation of Cassava Leaves by Two-Dimensional Gel Electrophoresis

摘要 通过优化等电聚焦强度改良木薯叶片蛋白质双向电泳的分离效果。结果表明,13万Vhs等电聚焦强度能很好地分离木薯叶片蛋白质,且分离的蛋白质具有良好的质谱兼容性。质谱鉴定7个蛋白点,结果显示这些蛋白主要参与能量代谢,其中有些蛋白点是在13万Vhs聚焦强度下特异出现的。 To obtain high-resolution reference of two-dimensional gel electrophoresis (2-DE)profiles of proteins from cassava leaves, the parameters of isoelectro-focusing(IEF)intensity were optimized. Results showed that the quality of 2-DE profiles under 130 000 Vhs was better than that of those under other conditions. Seven proteins among the selected 10 proteins were positively identified by MALDI TOF MS, suggesting their MS compatibility. The identified proteins were mainly involved in energy metabolic pathway and some of which appeared only in the 2-DE profiles under 130000 Vhs condition.
