欧李叶片过氧化物酶同工酶与叶果生理性状的相关性 被引量:6

摘要 为给欧李同工酶在数量性状遗传变异上的应用提供理论参考,利用聚丙烯酰胺凝胶电泳方法,对113份同一生境条件下野生欧李实生群体材料的叶片进行了过氧化物酶(POD)同工酶分析,同时对同工酶酶量大小(酶带宽窄)与叶片和果实生理性状的相关性进行了分析。研究结果表明:欧李实生群体叶片的过氧化物酶同工酶共分为7种酶谱类型,谱带数量最多能分离5条,其中Rf 0.126和Rf 0.716酶带为参试材料所共有的特征带,由Rf 0.126和Rf 0.716酶带组成的酶谱类型占参试材料总数的54.87%。系统聚类分析结果表明:同一生境条件下野生欧李实生群体单株个体间存在一定的遗传差异和亲缘关系;不同酶谱类型相同位置酶带不同单株个体间相对酶量的大小存在明显的差异,变异程度较大,同时欧李叶片和果实遗传性状表现差异明显。相关分析结果表明:不同酶谱类型中相关酶带相对酶量的大小与叶片和果实中相关生理性状指标存在着显著或极显著的相关性,这表明欧李实生群体过氧化物酶同工酶酶量大小的变化能够反映生物遗传性状的差异。 In order to provide reference for application of isoenzyme in researches on quantitative characters genetic variation,the peroxidase(POD) isoenzyme in leaves of 113 Cerasus humilis seedling strains which were cultivated at the same conditions were studied by the method of vertical slap polyacrylamide gel electrophoresis(PAGE),and the correlation of POD isoenzyme amount(band width) and physiological charateristics of leaves and fruits were analyzed.The results showed that 7 types of POD isoenzyme bands appeared in C.humilis seedling population,and the maximum number of bands was 5.Among thoes bands,two characteristic bands(Rf=0.126,Rf=0.716) appeared in all tested materials,and 54.87% of the tested materials just had the two bands.The results of system clustering analysis showed that the genetic difference and relationship existed among the individuals of C.humilis seedling population,the relative amounts of isoenzyme bands at the same position in different types of POD isoenzyme bands was different,and the variation was greater.Meanwhile,the genetic charateristics of leaves and fruits in C.humilis had obvious differences.The results of correlationship analysis showed that the notable or extremely notable relationship existed between amount of correlative isoenzyme bands in different types and the related physiological charateristic indexes,so amount of POD isoenzyme could indicate biological genetic charateristic difference in C.humilis seedling population.
