摘要
目的观察近紫外线对兔晶状体上皮细胞通讯功能的影响,探讨星状孢菌素在近紫外线辐射时对细胞通讯功能的保护作用。方法建立体外培养兔晶状体上皮细胞的近紫外线辐射模型,星状孢菌素作为实验因素,设为正常组、对照组和实验组,应用激光共聚焦显微镜荧光光漂白恢复技术检测细胞的荧光恢复率以评估细胞的通讯功能,采用免疫印迹法分析各组连接蛋白(Cx)43的表达水平。结果近紫外线照射15min后,实验组及对照组的细胞形态均发生变化,但实验组的变化较对照组轻。近紫外线照射10和15min后,实验组荧光恢复率均显著高于对照组(P值均<0.05)。近紫外线照射10min后,实验组晶状体上皮细胞Cx43的表达显著高于对照组(P<0.05),但显著低于正常组(P<0.05)。结论近紫外线辐射可损伤晶状体上皮细胞的通讯功能,星状孢菌素对细胞通讯功能具有显著的保护作用。
Objective To observe the influence of near ultraviolet ray on intercellular communication in rabbit lens epithelial cells and to study the protective effects of Staurosporine on intercellular communication affected by near ultraviolet ray.Methods Rabbit lens epithelial cells cultured in vitro were exposed to near ultraviolet ray,and Staurosporine was added into the cell culture liquid.Fluorescence recovery after photobleaching(FRAP) was used to determine the fluorescence recovery rate and intercellular communication,and the expression level of Connexin 43(Cx43) was detected by Western blotting analysis.Results The morphology of epithelial cells changed in both treatment group(ultraviolet irradiation + Staurosporine) and control group(ultraviolet irradiation without Staurosporine) 15 minutes after near ultraviolet irradiation,but the change in treatment group was smaller than that in control group.The fluorescence recovery rate was significantly higher in treatment group than that in control group 10 and 15 minutes after near ultraviolet irradiation(all P0.05).The expression level of Cx43 in treatment group was significantly higher than that in control group,and lower than that in normal group 10 min after near ultraviolet irradiation(all P0.05).Conclusion Near ultraviolet irradiation can destroy the intercellular communication in rabbit lens epithelial cells,while Staurosporine can significantly protect the intercellular communication.
