葛根异黄酮对AD模型大鼠抗衰老作用研究 被引量:6

摘要 目的研究葛根异黄酮对D-半乳糖致衰老大鼠的抗衰老作用。方法 wistar大鼠68只,随机分为对照组、模型组、药物组低、高剂量组,每组17只。造模成功后,第二天开始灌胃给药,药物低、高剂量组每天分别给予40 mg/kg、160 mg/kg葛根异黄酮灌胃,模型组、对照组每天给予80 mg/kg生理盐水灌胃,连续8周后取血液,并将大鼠断头处死后取肝脏及脑组织,制作海马10%组织匀浆,分别检测丙二醛(MDA)及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、单胺氧化酶(MAO)水平的变化情况。结果模型组大鼠血液、肝脏、海马中SOD、GSH-Px的活性明显降低,MDA的含量明显增加,与对照组相比有明显差异(P<0.01);与模型组相比,药物低、高剂量组大鼠,除MAO外各项指标明显好转(P<0.05),且高剂量组比低剂量组的作用更明显(P<0.01);血液、肝脏组织中MAO的活力用药后,低、高剂量组均未见显著性改变,而在海马组织中有显著性降低(P<0.05)。结论葛根异黄酮能增加血液和肝脏中SOD、GSH-Px活力,降低血液、肝脏和脑组织中MDA的含量,具有一定的抗衰老作用。 Objective To investigate the anti-aging effects of Pueraria isoflavones on AD model rats.Methods 68 wistar rats were randomly divided into control group,model group and medication groups with low and high dose averagely.After a successful modeling,began the next day gavage,medication groups with low and high dose were given 40mg/kg and 160mg/kg isoflavones puerarin respectively daily,while model group and control group were given 80mg/kg normal saline daily.After 8 weeks,blood was drawn,and the liver and brain tissue of rats were taken after decapitation to produce 10% of hippocampal tissue so as to measure the enzymatic activity of malondialdehyde(MDA) and superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),monoamine oxidase(MAO).Results The activity of blood,liver,hippocampus SOD and GSH-Px in the model group were significantly decreased,while MDA level increased significantly.Compared with the control group,there was significant difference(P0.01).Compared with model group,medication groups with low and high dose showed significant improvement in above parameters(P0.05),and the high-dose group showed better results than low-dose group(P0.01);after treatment,blood and liver tissue MAO vitality in both low and high dose groups showed no significant changes,but it was significantly lower in the hippocampus(P0.05).Conclusion Pueraria isoflavones has anti-aging effects in that it can increase the SOD and GSH-Px activity of i blood and liver and lower the MDA activity of blood,liver and brain tissue.
