实验性慢性萎缩性胃炎脾气虚证模型的建立及不同时期病理形态学的改变

Establishment of Spleen Qi Deficiency Syndrome of Experimental Chronic Atrophic Gastritis Model And Pathological Changes in Different Stages

目的:采用N-甲基-N-硝基-N-亚硝基胍(MNNG)和40%乙醇灌胃建立大鼠慢性萎缩性胃炎(CAG)模型,同时采用破气苦降,饥饱失常和疲劳的方法进一步建立CAG脾气虚证大鼠模型。观察CAG脾气虚证大鼠不同时期病理形态学的改变。方法:以170μg/mL的MNNG10mL/kg灌胃,150μg/mL的MNNG溶液给大鼠自由饮用,40%乙醇,2mL/只,每周2次,在此基础上第5周采用破气苦降,饥饱失常和疲劳的方法建立CAG脾气虚证大鼠模型,连续10周。每2周随机处死CAG脾气虚证大鼠2只进行光镜观察。结果:随着造模时间的延长动物胃部病变的进程是从正常胃黏膜-慢性胃炎-萎缩性胃炎-肠上皮化生-不典型增生-胃癌前病变。结论:采用MNNG和40%乙醇灌胃以及破气苦降,饥饱失常和疲劳的方法可成功建立慢性萎缩性胃炎脾气虚证模型。

Objective：Chronic atrophic gastritis rat model was induced by intragastric administration of MNNG and 40% ethanol. Furthermore, spleen deficiency syndrome of the CAG model was established by irregular feeding and overstrain. Pathological changes in different stages were observed. Methods ：Intragastric administration of MNNG with the dose of 10mL/kg（ 170μg/mL）was given and MNNG （150μg/mL） was for drinking. 40% ethanol was injected twice a week with the dose of 2mL for every rat. Besides, in the 5th week, spleen deficiency model was established by irregular feeding and overstrain continuously lasting 10 weeks. Every 2 weeks, two model rats were put to death randomly for light mi- croscope observation. Results： With the time passing by, the development of the CAG was as following：normal gastric mucosa - chronic gastritis - atrophic gastritis - intestinal metaplasia - atypical hyperplasia - gastric precancerous lesion. Conclusion：MNNG and 40% ethanol with irregular feeding and overstrain can successfully establish the spleen deficiency syndrome of chronic atrophic gastritis model.
