电针不同穴位对心肌肥厚模型大鼠炎症细胞因子的比较研究 被引量:5

摘要 目的:观察电针不同穴位对心肌肥厚模型大鼠炎症细胞因子的影响,探讨不同穴位对心肌肥厚模型大鼠的保护作用的差异。方法:SD大鼠50只随机数字表法分为正常组、模型组、"内关"组、"合谷"组、"神门"组,采用皮下注射盐酸异丙肾上腺素3mg.kg-1.d-1建立心肌肥厚大鼠模型,电针组采用连续波,频率2Hz,强度1mA,通电20min,1次/天,共14天。测定左室壁厚度和心肌细胞直径,放射免疫法技术检测心肌组织和血清TNF-α和IL-1β含量,研究电针"内关"、"合谷"、"神门"穴对左室壁厚度、心肌细胞直径、TNF-α、IL-1β的影响,并对数据进行统计分析。结果:模型组大鼠与正常组大鼠比较,左室壁厚度、心肌细胞直径、TNF-α和IL-1β显著升高(P<0.01),"合谷"组、"内关"组、"神门"组大鼠左心室壁平均厚度、心肌细胞平均直径均明显低于模型组(P<0.05,P<0.01),3组之间比较"合谷"组优于"内关"组、"神门"组(P<0.05,P<0.01),"内关"组、"神门"组之间无显著性差异(P>0.05);"内关"组、"合谷"组、"神门"组大鼠TNF-α、IL-1β均明显低于模型组(P<0.05),3组穴位之间比较无显著性差异(P>0.05)。结论:电针"内关"、"合谷"、"神门"可以有效防治心肌肥厚,"合谷"优于"内关"、"神门",这种保护作用可能与抑制TNF-a、IL-1β等炎症细胞因子的表达,抑制炎症反应有关,抑制炎症反应可能是其抗心肌肥厚作用机制之一。 Objective : To discuss the influence of electroacupuncture at Neiguan, Hegu and shenmen on the inflammatory cytokines in rats with cardiac hypertrophy. Methods: Fifty SD rats were randomly divided into five groups by random number table method : the normal group, the model group, the Neiguan group, the Hegu group, the Shenmen group, with ten rats in each group respectively. Rats in model group were received 3mg · kg-1 · d-1 of isoprenaline hydrochloride, the electroacupuncture group selected a stimulus parameter: continuous -wave, frequency of 2HZ, current intensity of 1 mA, and treated for twenty minutes, once a day, amounted to 14 days;We measured the thickness of left ventricular wall and the diameter of the cadiocyte through pathological examination, the contents of TNF - α and IL - 1β of heart tissue and blood plasma were determined by radioimmnnoassay, then analysis the data through an statistical analysis system. Results: Compared with the normal group, the thickness of left ventricular wall , the diameter of the cadiocyte , the contents of TNF - α and IL - 1β in heart tissue and blood plasma of the rats were significantly higher with statistical signifi- cance in normal group ( P 〈 0.01 ) ; compared with the model group, the thickness of left ventricular wall and the diameter of the cadiocyte were lower with statistical significance in the Neiguan group, the Hegu group, the Shenmen group( P 〈 0.05, P 〈 0. 01 ) ;the Hhegu group is better than the Neiguan group and the Shenmen group( P 〈 0.05, P 〈 0.01 ), there is no difference between the Neiguan group and the Shenmen group ( P 〉 0.05). Compared with the model group, contents of TNF - α and IL - 1β in heart tissue and blood plasma of the rats were significantly lower with statistical significance in the Neiguan group, the Hegu group, the Shenmen group ( P 〈 0. 05 ), but no significant difference among them( P 〉 0. 05 ). Conclusion: Electroacupuncture at Neiguan, Hegu and Shenmen can prevent the myoeardial hypertrophy caused by injection with isoprenaline hydrochloride, the Hegu was superior than Neiguan and Shenmen,which were associated with its anti - inflammation effect, there is no difference among them on inflammatory cytokines.
