摘要
研究中药大黄三七散对急性脑出血(ICH)大鼠的神经功能缺损改善的影响并探讨其分子机制。以肝素胶原酶Ⅳ法制作大鼠脑出血模型,考察了施药前后ICH大鼠神经功能评分、脑系数及脑含水量、脑匀浆中自由基释放量及抗氧化酶活力的变化及病理切片观察。结果表明,大黄三七散可明显改善ICH大鼠神经功能缺失症状,显著降低脑系数并减少脑含水量,施加大黄三七散后ICH大鼠脑组织自由基含量明显降低,而机体SOD及CAT抗氧化酶活力提高。改善效果明显呈剂量依赖效应。病理切片观察表明,与模型组相比,大黄三七治疗3个剂量治疗组ICH大鼠的脑组织病变、脑水肿及炎症细胞浸润情况均明显改观,神经细胞损伤亦显著减轻。大黄三七散的解毒、通络及抗氧化功效是改善急性ICH大鼠神经功能缺损的重要分子机制。
This paper was to study the improvement of Dahuangsanqi powder on the neural function deficiency in rats with intracerebral hemorrhage (ICH) and explore its mechanism. The ICH model was established by the collagen enzymes IV infusion method. Neurologieal scale, cerebral indexes, water content, the NO level, iNOS,CAT,SOD and LPS activities in cerebral tissue were measuerd in addtion to the observation of the brain histological changes. The results showed that adminstration of the Dahuangsanqi powder can obviously improve the neurologieal scale, decrease the cerebral indexes and water contents and the induce free radicals levels, and improve the SOD and CAT activities in dose - dependent manners. The treatment of three doses groups of Dahuangsanqi powder can significantly improve the histological changes and reduce the nerve cells damage. In conclustion, we indicated that the abilities of detoxifcation and collateral dredge and antioxidant capacity of Dahuangsanqi powder was one of the important aspect for the mechanisms for its improvement on neural function deficiency in rats with ICH.
