摘要
目的:探讨结核分枝杆菌(Mycobacterium tuberculosis,Mtb)感染THP-1细胞后对细胞表面白细胞分化抗原14(CD14)表达的影响。方法:Mtb以10∶1感染佛波醇酯(PMA)分化的THP-1细胞,建立Mtb感染THP-1细胞的模型。采用流式细胞术检测THP-1细胞、PMA分化的THP-1细胞及Mtb感染THP-1细胞CD14的表达。结果:THP-1细胞和PMA分化的THP-1细胞表面表达CD14的阳性率分别为5.5%和21.6%,对Mtb的吞噬率分别为7.7%和93.0%;Mtb感染的THP-1细胞表面表达CD14的阳性率为27.5%。结论:PMA分化后的THP-1细胞表面CD14的表达增加,Mtb感染后CD14的表达进一步增加,对Mtb的吞噬率增加。
Objective: To explore the changes of CD14 expression before and after THP-1 cells infected with Mycobacterium tuberculosis(Mtb). Methods: Mtb and phorbol myristate acetate (PMA)-differentiated THP-1 cells were incubated with the ratio of 10:1 ,and Mtb infection cell model was established. The CD14 expression in THP-1 cells, PMA differentiated THP-1 cells and Mtb infected THP-1 cells were detected by flow cytometry. Results: The percentage of CD14 expression in THP-1 ceils and PMA differentiated THP-1 cells was 5.5%, and 21.6%, respectively, whereas the percentage of Mtb phagocytosis rate was 7.7% and 93.0%, respectively. The percentage of CDI4 expression in Mtb infected THP-1 cells was 27.5%. Conclusions:The CDI4 expression in PMA differentiated THP-1 ceils is increased, and the CD14 expression and Mtb phagocytosis rate in Mtb infected THP-1 cells are further increased.
