缝隙连接:一个潜在的肿瘤治疗新靶点 被引量:5

摘要 连接蛋白(connexin,Cxs)及其形成的缝隙连接细胞间通讯(gapjunctioncell—cellcommunication,GJIC)在肿瘤的生长、增殖、转移中起着重要的作用。研究[1]证实,在培养的正常细胞和肿瘤细胞,缝隙连接(gapjunction,GJ)能够增强多种化学毒物引起细胞凋亡的作用。大量研究[2]表明,靶向Cxs和GJIc的治疗可作为一个崭新的技术直接抑制肿瘤生长或使其他的治疗方法更敏感。本文就GJ和Cxs杀灭肿瘤细胞的各种方法,包括旁观者效应(bystandereffect,BE)基因治疗,转录与转录后水平,恢复Cxs表达的常用方法,以及该治疗方法的可行性和局限性作一综述。
