摘要
目的探讨动脉粥样硬化闭塞症(ASO)兔模型TNF-α、NF-κB mRNA和蛋白表达的变化,部分阐明ASO炎症反应机制。方法建立ASO动物模型,以免疫组化法观察主动脉内皮组织TNF-α、NF-κB蛋白表达,RT-PCR方法观察TNF-αmRNA、NF-κBmRNA表达情况。通过免疫组化指数以及目的基因的相对转录量为参数进行统计分析。结果 ASO模型兔主动脉内膜明显增厚,斑块面积明显增加。模型组兔主动脉内皮组织TNF-α、NF-κB蛋白表达和TNF-αmRNA、NF-κBmRNA的表达均较对照组升高(P<0.01)。结论 ASO发生发展与炎症反应有关。
Objective To observe the level changes of TNF-α 、NF-κB and TNF-αmRNA、NF-κBmRNA in rabbit artery with the atherosclerotic occlusive disease(ASO).Methods ASO model was established in rabbits.Changes of TNF-α 、NF-κB were detected by the Immunohistochemistry and the changes of TNF-αmRNA、NF-κBmRNA were monitored by the reverse transcrip tase polymerase chain reaction assay.Results Compared the model group with the control group,the expression level of TNF-αmRNA、NF-κBmRNA showed significant increase(P0.01).Conclusion The level of TNF-α、NF-κB and TNF-αmRNA、NF-κBmRNA were high expression on the established process of the atherosclerotic occlusive disease(ASO.The mechanism of inflammation is closely related to the atherosclerotic occlusive disease.
