乳糖诱导表达重组人甲状旁腺激素rhPTH(1-34)

摘要 目的研究以乳糖代替异丙基硫代半乳糖苷(IPTG)诱导表达重组人甲状旁腺激素的可行性。方法对乳糖浓度、诱导时间及发酵培养方式进行研究,确定了乳糖诱导的最佳条件。结果 0.6%的乳糖诱导4 h,目的蛋白的表达量约占菌体总蛋白的56.8%,采取分批补料培养方式,5 L发酵罐中蛋白表达量达到42.6%。结论乳糖能作为诱导剂诱导rhPTH(1-34)在大肠杆菌中的表达。 Objective To investigate the feasibility of expression of recombinant human parathyroid hormone induced by lac- tose instead of IPTG. Methods The influences of inducer concentrationt induction time and culture method of fermentation were investigated to determine the optimal induction conditions. Results The target protein was about 56.8% of total cellular proteins with 0.6% lactose at 4 h, and the expression level reached about 42.6% in 5 L fermentor with fed - batch culture. Conclusion Lactose could be used as inducer in the expression of rhPTH ( 1 - 34 ) in Escherichia coli.
