摘要
目的应用高速逆流色谱法(HSCCC)分离望春花蕾中的木兰脂素。方法以石油醚-乙酸乙酯-甲醇-水(5∶2∶3∶6,v/v)为两相溶剂体系,以下相为流动相,在流速为2.0 mL.min-1,主机转速为800 r.min-1,检测波长为254nm条件下进行分离。结果从180 mg提取物中分离得到30 mg木兰脂素,通过电喷雾电离质谱(ESI-MS)和核磁共振波谱(NMR)对其结构进行了鉴定。结论本方法重现性好,适合木兰脂素的制备性分离。
Objective To separate magnolin from flower buds of magnolia biondii by high - speed counter current chroma- tography ( HSCCC ). Methods The separation was performed with a two - phase solvent system composed of petroleum ether - ethyl acetate - methanol - water ( 5 : 2 : 3 : 6, v/v ). The lower phase was used as the mobile phase with a flow rate of 2.0 mL · min^- 1, the apparatus rotated at 800 r · min^- 1 and the effluents were detected at 254 nm. Results 30 mg magnolin was separated from 180 mg extract ,and the chemical structure of magnolin was identified by electrospray ionization mass spectrometry ( ESI - MS) and nuclear magnetic resonance spectroscopy ( NMR ). Conclusion This method was reproducible and suitable for the separation and preparation of magnolin.
