IL-17在小鼠变应性鼻炎模型中作用的实验研究 被引量:1

Effect of IL-17 in Murine Model of Allergic Rhinitis

摘要 目的研究IL-17与变应性鼻炎(allergicrhinitis,AR)的关系。方法选取6~8周Balb/c雄性小鼠36只随机分组模型组、IL-17抗体干预组、对照组,每组12只。利用鸡卵清白蛋白(OVA)建立Balb/c小鼠变应性鼻炎模型。于激发期记录症状表现。末次激发后处理动物,取鼻腔固定脱钙后染色,观察其形态学特点,取脾脏获取淋巴细胞,提取RNA行逆转录-聚合酶链反应(reversetranscnption-polymerasechain reaction,RT-PCR),检测IL-17 mDNA、IL-5 mDNA、IFN-γmRNA的表达情况。结果与其他两组比较,干预组的变应性鼻炎症状加重,鼻腔粘膜嗜酸性粒细胞浸润现象加重。IL-5 mRNA的相对表达水平也要高于模型组与对照组(P<0.05),而对照组IL-17 mRNA相对表达水平要高于模型组与干预组(P<0.05)。结论 IL-17在变应性鼻炎中通过抑制辅助性T细胞2(Th2)细胞的功能,减轻免疫应答的方式起到一定的调节作用。 Objectlve To find out the correlation between IL-17 and allergic rhinitis (AR). Methods 36 male BALB/c mice were divided into 3 groups: ovalbumin(OVA) group, intervention group and controls. The BALB/c mice allergic rhinitis models was induced by ovalbumin (OVA). The symptoms were observed in challenge phase. After last challenge, the pathological differences between the three groups were be surveyed. The levels of IL-17 mRNA ,IL-5 mRNA and IFN-~/mRNA in lymphocytes from spleen were measured by reverse transcription-poly- merase chain reaction (RT-PCR). Results In contrast to the others, intervention group had significant eosinophil infiltration in the nasal cavity. The sever symptoms of allergic rhinitis were observed in intervention group. IL-5 mRNA expression of intervention group were higher than those in the others (P 〈 0. 05 ). IL-17 mRNA expression of controls were higher than those in the others (P 〈 0. 05 ). Conclusion IL-17 plays an important regulatory role in allergic rhinitis by inhibiting the allergic response and the function of help T cell 2 (Th2).
