摘要
目的通过建立两次打击大鼠致多器官功能障碍综合征(MODS)的动物模型,探讨黄芪注射液对MODS致血管内皮细胞损伤的影响。方法将86只SD大鼠随机分为对照组、MODS组和治疗组。MODS组和治疗组复制大鼠MODS的动物模型,治疗组腹腔注入黄芪注射液,对照组和MODS组注入0.9%氯化钠溶液。分别在注射黄芪注射液和0.9%氯化钠溶液后24和48 h采集血液标本,测定大鼠血清中循环内皮细胞(CEC)数量、血栓调节蛋白(TM)、血管性血友病因子(VWF)、细胞间黏附分子-1(ICAM-1)、超氧化物歧化酶(SOD)、丙二醇(MDA)的含量。结果治疗组大鼠腹腔注射黄芪注射液后24和48 h CEC数量、TM、VWF含量明显低于相同时间点MODS组(P<0.05);治疗组大鼠腹腔注射黄芪注射液后24和48 h ICAM-1、MDA含量明显低于MODS组相同时间点含量(P<0.01),而24和48 h SOD含量治疗组明显高于MODS组(P<0.01)。结论黄芪注射液对两次打击致多脏器功能衰竭模型大鼠的血管内皮细胞损伤有明显的保护作用。
[ Objective ] To study the effects of astragalus injection on vascular endothelial cells injury in rat models of multiple organ dysfunction syndrome (MODS) induced by harmful two-hit. [ Methods ] Eighty-six SD rats were randomly divided into control group, MODS group and treated group. MODS models were reproduced in MODS and treated groups. Astragalus solution was injected in treated group, meanwhile control and MODS groups were in- jected with 0.9% saline. Blood specimens were collected at 24 h, 48 h, respectively after injection to determine number of circulating endothelial cells (CEC), contents of thrombomodulin (TM), yon Willebrand factor (VWF), intercellular adhesion molecule-1 (ICAM-1), superoxide dismutase (SOD) and malondialdehyde (MDA). [ Results] Number of CEC, contents of TM and VWF at 24 h and 48 h in treated group were lower obviously than in MODS group. at same time points (P 〈0.05). Contents of ICAM-1 and MDA at 24 h and 48 h in treated group were lower obviously than in MODS group at same time points (P 〈0.01), contents of SOD at 24 h and 48 h in treated group were higher obviously than in MODS group at same time points (P 〈0.01). [ Conclusion] Astragalus injection can protect vascular endothelial cells injury in rat models of MODS induced by two-hit.
