摘要
目的检测胃癌组织及胃癌细胞中微小RNA(miRNA)表达谱,并测定胃癌组织及胃癌细胞中miR-429的表达水平。方法采用基因芯片技术检测胃癌组织、癌旁正常胃组织标本及胃癌细胞株(SGC-7901)、正常人胃上皮细胞(GES-1)中miRNAs的表达。挑选基因芯片结果中胃癌组织表达显著下调的miR-429,并采用实时定量聚合酶链反应(qRT-PCR)检测7例正常胃组织、52例胃癌组织、胃癌细胞SGC-7901、GES-1中miR-429的表达。结果基因芯片结果表明,胃癌组织中有23个miRNA上调,如miR-21、miR-30b等,31个下调如miR-429、miR-200等。qRT—PCR检测显示,miR-429在胃癌组织中下降约63.4%(P〈0.01),在胃癌细胞株SGC7901中下降约76.2%(P〈0.01)。结论miR-429在胃癌组织中表达显著下调,可能作为胃癌特异性miRNA,在胃癌的发生发展过程中有重要作用。
Objective To detect the expression profiling of microRNA (miRNA) and the expres- sion level of miRNA-429 in human gastric carcinoma. Methods The miRNAs expression of 7 normal gas- tric tissues, 52 malignant tissues, gastric cancer cell line SGC7901 and gastric epithelial cell line 1 ( GES-I ) was detected by using miRNA gene chips. Real-time reverse transcriptase-polymerase chain reac- tion (qRT-PCR) was used to quantify the expression level of miR-429 in 52 gastric cancer tissues and their paracancerous tissues. Results The results of gene chip showed that 23 miRNA were significantly up-regu- lated, such as miR-21, miR-30b, etc. , and 31 miRNA down-regulated, such as miR429, miR-200, etc. The average expression of miR-d29 was significantly reduced by 63.4% in gastric cancer samples as com- pared with that in normal paracancerous tissues (P 〈 0. 01 ). miR429 expression was reduced by 76. 2% in SGC-7901 cells as compared with non-tumor gastric cells (P 〈 0. 01 ). Conclusion miR429 might play a role in the pathogenesis of gastric carcinoma.
