摘要
目的观察胃转流术(GBP)对糖尿病大鼠血糖的控制效果及胰高血糖素样肽-1(GLP-1)的影响。方法采用链脲佐菌素建立糖尿病sD大鼠模型20只,随机分为糖尿病手术组(DO组)和糖尿病对照组(DC组),另取20只非糖尿病大鼠随机分为正常对照组(NC组)和正常手术组(NO组)。分别检测各组大鼠术前、术后72h、1周、4周和8周空腹血糖水平以及血清GLP-1浓度。结果术前DO组与DC组以及NC组与NO组大鼠空腹血糖之间的比较差异均无统计学意义(P〉0.05);DO组大鼠术后空腹血糖进行性下降,术后8周由术前的(20.84±1.98)mmol/L下降到(5.56±0.11)mmol/L(P〈0.05);DC组大鼠术前及术后各时相的差异无统计学意义(P〉0.05)。DO组和NO组大鼠术后血清GLP.1浓度出现明显升高(P〈0.05),术后8周分别由术前的(7.10±0.55)、(10.73±0.67)pmol/L上升到(26.48±1.14)、(13.98±0.92)pmol/L(P〈0.05)。结论GBP对2型糖尿病大鼠具有明显的降糖作用,GLP.1的升高在其中起着重要作用,但对正常大鼠血糖无影响。
Objective To investigate the effect of gastric bypass surgery (GBP) on fasting blood- glucose (FBG) and glucagon-like peptide-1 (GLP-1) in type 2 diabetic rats. Methods The models of type 2 diabetic rats were induced by stretozotocin and 20 diabetic rats were randomly divided into two groups: diabetes-operation group (DO group, n = 10) and diabetes-control group (DC group, n = 10). Another twenty normal rats were randomly divided into two groups: normal-operation (NO group, n = 10) and normal-control group ( NC group, n = 10). The FBG levels and glucagon-like peptide-1 ( GLP-1 ) con- centrations of rats in each group were detected before operation and 72 h, on 1 week, 4 weeks, 8 weeks af- ter operation. Results The FBG levels of rats before operation were not statistically significant different between DO group and DC group or between NO group and NC group (P 〉 0. 05). After operation, the FBG levels of rats in DO group gradually declined, eight weeks after operation the FBG levels was declined from (20. 84 ± 1.98) mmol/L to (5. 56± 0. 11 ) mmol/L (P 〈 0. 05) ; In DC group, the difference of FBG levels of rats at different time point was not statistically significant (P 〉 0. 05). After operation, the con- centrations of serum GLP-1 of rats in DO group and NO group gradually increased, eight weeks after operation The serum GLP-1 elevated respectively from ( 7. 10 ± 0. 55 ), ( 10. 73 ± 0. 67 ) pmol/L to ( 26. 48 ± 1.14), (13.98 ±0. 92) pmol/L (P 〈0. 05). Conclusion There is obvious hypoglyeemic effect of GBP on FBG levels of type 2 diabetic rats other than normal rats, in which high secretion of GLP-1 may be play an important role, high secretion of GLP-1 on the normal type 2 diabetic rats has no hypoglycemie effect.
