骨髓基质细胞源内皮细胞移植对血管性痴呆大鼠行为及VEGF蛋白表达的影响

Effects of bone marrow stromal cells derived endothelial cells on the dynamic changes of behavior and expression of VEGF protein in vascular dementia rats

摘要 目的 探讨骨髓基质细胞(BMSCs)向血管内皮细胞诱导分化后移植对血管性痴呆(VD)大鼠行为及血管内皮生长因子(VEGF)蛋白表达的影响.方法 采用2-AO法制作VD大鼠模型;经Y型电迷宫筛选健康SD大鼠随机分为假手术组(n=8)、PBS移植组(n=8)、BMSCs移植组(n=8)、BMSCs源内皮细胞移植组(VECs移植组,n=8);移植30天后Y型电迷宫检测大鼠学习记忆能力;处死大鼠,大鼠脑切片检测VEGF蛋白表达、海马CA1区锥体细胞数.结果 VECs移植组、BMSCs移植组、PBS移植组大鼠学习记忆能力均较假手术组下降(P〈0.05).移植后30天各组神经功能均有不同程度的恢复,VECs移植组大鼠的学习记忆能力显著优于BMSCs移植组(P〈0.05);VECs移植组VEGF阳性细胞多于BMSCs移植组(P〈0.05);VECs移植组海马CA1区锥体细胞数高于BMSCs移植组(P〈0.05).结论 BMSCs源内皮细胞移植町改善VD大鼠学习记忆能力,增加VEGF蛋白表达和保护海马区神经元,显著优于BMSCs移植. Objective TO study the effects of bone marrow stromal cells (BMSCs) derived endothelial cells on behavior and expression of vascular endothelial growth factor (VEGF) protein in vascular dementia (VD) rats. Methods The model of VD was established by 2 - AO. 32 healthy Sprague - Dawley rats selected by Y - Morris electric maze experiment were divided into sham - operated group ( n = 8), PBS transplanted group ( n = 8), BMSCs transplanted group ( n = 8) , and BMSCs derived endothelial ceils transplanted group ( VECs transplanted group, n = 8 ). The learning and memory ability of the rats were evaluated 30 days after transplantation. The rats were killed separately 30 days after opera- tion. The brain sections were used for detecting the expression of the VEGF protein and the number pyramidal neurons in the hippocampal CA~. Results The learning and memory ability of VECs transplanted group, BMSCs transplanted group and PBS group was lower than that of sham - operated group ( P 〈 O. 05 ). The recovery of learning and memory ability in VECs transplanted group was evidently better than BMSCs transplanted group 30 days after transplantation ( P 〈 0.05 ). The number of VEGF positive cells in VECs transplanted group increased evidently compared with that of the BMSCs transplanted group ( P 〈 O. 05 ) . The number of pyramidal neurons in the hippocampal CAI was significantly higher in VECs transplanted group than in BMSCs group ( P 〈 0.05 ). Conclusion Compared with BMSCs, BMSCs derived endo- thelial cells are better in improving the learning and memory ability, improving the expression of VEGF protein and reducing the depletion of pyramidal neurons in the hippocampal CAi in VD rats.
