热疗对膀胱癌细胞BIU-87HCPT耐药相关基因的影响

摘要 目的探究热疗对膀胱癌细胞BIU-87HCPT耐药相关基因表达的影响。方法培养试验用对羟基喜树碱耐药的膀胱癌细胞株BIU-87HCPT,待其处于生长对数期时取出待用。将对照组与实验组均放置于37~43℃不同的温度的恒温箱中化疗,72 h后用酶标仪测量其吸光度。结果单独热疗可以抑制BIU-87/HCPT癌细胞,且抑制效果与温度呈正相关。热疗联合化疗比单独热疗或者单独化疗效果好。热疗可抑制BIU-87/HCPT癌细胞繁殖。热疗可抑制P-gp、MRP1、GST-π和survivin的表达,且抑制效果与温度呈正相关。结论热疗可以很好的抑制膀胱BIU-87/HCPT肿瘤细胞的生长繁殖,抑制其表达耐药相关蛋白从而降低其耐药性以提高疗效。 [ Objectives ] To explore the effects of hyperthermia on the resistance related gene of bladder cancer cell. [ Methods] Bladder cancer cell line BIU-87HCPT which was resistant to hydroxycamptothecin were cultured to prepare for the experiment when it reached the logarithmic growth phase. The control group and experiment group were both placed in the incubator with 37-43℃, and the absorbance were tested with enzyme mark instrument 72 hours later. [ Results ] The cancer cells BIU-87/HCPT could be inhibited by hyperthermia alone, and the inhibition effect was positively correlated with the temperature. The effect of combined treatment of hyperthermia and chemotherapy was better than that of hyperthermia alone. The propagation of the bladder cancer cell BIU-87HCPT could be inhibited by hyperthermia. The expression of the P-gp, MRP1, GST-π, survivin could also be inhibited by hyperthermia, and the inhibition effect was positively correlated with the temperature. [ Conclusion ] The growth and reproduction of the bladder cancer cell BIU-87HCPT can be inhibited well by hyperthermia, and so can the expression of the resistance related protein to decrease the resistance and improve the effect of the treatment.
