胰腺癌高淋巴转移细胞系的建立及其肿瘤干细胞生物学特性的初步研究 被引量:4

摘要 背景与目的:胰腺癌淋巴道转移特性研究越来越受到重视,构建胰腺癌淋巴道转移动物模型能够为相关研究提供理想的实验平台。建立一种具有淋巴道高转移活性的人胰腺癌细胞系,可为深入研究胰腺癌淋巴转移的机理,预防和治疗胰腺癌淋巴转移提供合适的实验平台。方法:通过将人胰腺癌细胞株BxPC-3裸鼠爪垫皮下连续接种法建立淋巴道转移模型,筛选、建立高淋巴转移胰腺癌细胞系,采用流式细胞术检测细胞系中CD133的表达。结果:成功构建了裸鼠人胰腺癌细胞株BxPC-3淋巴道转移模型,获得了具有高侵袭性、高淋巴转移能力的胰腺癌BxPC-3-LN细胞系。该细胞系具有侵袭力强、转移效率高和转移集中的特点,在胰腺原位和爪垫的成瘤能力明显强于母系BxPC-3细胞,其淋巴结转移的发生率明显高于母代细胞。BxPC-3-LN细胞系CD133阳性细胞比例(3%)明显高于母代细胞(0.5%)。结论:该细胞系为胰腺癌转移机理的研究和抗肿瘤药物的筛选提供了合适的实验平台,具有非常重要的科学意义和应用前景。 Background and purpose:The molecular mechanisms of lymph node involvement has been investigated extensively.Animal model plays an important role in these research.The purpose of this study was to establish a highly lymph-metastasis pancreatic cancer cell line.Methods:An in vivo highly continuous selection method was employed.Flow cytometry was used to test the expression of CD133 positive cells.Results:A highly lymph-metastasis pancreatic cancer cell line,BxPC-3-LN,was established.Compared with the parental BxPC-3 cell line,the BxPC-3-LN cell line has higher migration and invasion potential and tumorgeniety.The rate of lymphatic metastasis of the BxPC-3-LN cell line(100%) was greatly higher than that of the parental BxPC-3 cell line(5%).The percentage of CD133 positive cells(3%) is higher than the parental cells(0.5%).Conclusion:This highly lymph-metastasis pancreatic cancer cell line will provide an appropriate and valuable platform to study the mechanism of the lymph-metastasis of pancreatic cancer.
