摘要
背景与目的:真核细胞翻译起始因子4E(eukaryotic initiation factor 4E,eIF4E)是蛋白翻译过程中的调控因子,血管内皮生长因子(vascular endothelial growth factor,VEGF)是血管生成过程中的关键因子,eIF4E及VEGF的表达异常可能参与肿瘤的发生、发展。本研究旨在探讨eIF4E及VEGF与子宫内膜癌发生、发展的关系及其临床意义。方法:采用免疫组化SP法检测eIF4E与VEGF在27例子宫内膜腺癌(endometrial adenocarcinoma,EA)、16例Ⅱ型子宫内膜癌及20例正常子宫内膜中的表达情况,并分析两者与子宫内膜癌临床病理特征的关系以及两者表达的相关性。结果:eIF4E在Ⅰ型及Ⅱ型子宫内膜癌中表达的MVD值分别为0.18±0.05和0.20±0.07(P=0.224),两组表达与正常组相比,差异均有统计学意义(P<0.000);VEGF在Ⅰ型及Ⅱ型子宫内膜癌表达的MVD值分别为0.16±0.06和0.18±0.07(P=0.387),两组与正常组相比,差异均有统计学意义(P<0.000)。eIF4E表达与子宫内膜腺癌FIGO分期、组织学分级及淋巴转移呈正相关(P<0.05),与年龄及浸润深度无关(P>0.05);VEGF表达与FIGO分期、组织学分级、淋巴转移及浸润深度呈正相关(P<0.05)。eIF4E及VEGF在子宫内膜癌中的表达呈正相关(r=0.565,P=0.001)。结论:eIF4E及VEGF与子宫内膜癌的发生、发展及生物学行为关系密切,可初步考虑作为子宫内膜癌恶性改变的分子标志。
Background and purpose: eIF4E is the regulation factor of protein translation process and VEGF is the key factor in angiogenesis. The abnormal expression of eIF4E and VEGF are correlated with the emergence and development of tumor. This study tested the expression of eIF4E and VEGF in endometrial carcinoma (EC) so as to investigate the clinical significance between them and EC. Methods: In 27 cases of endometrial adenocarcinoma (EA), 16 cases of EC (typeⅡ ) and 20 cases of normal endomedium (NE), the expressions of eIF4E and VEGF were detected by immunohistochemical method of SP and the relationship with the pathological phenotype of EA were analyzed. Results: The mean optical density (MOD) of eIF4E in EA and EC (type Ⅱ) were 0.178m0.053 and 0.201±0.067 (P=0.224), respectively, which were higher than that of NE (P〈0.000). The MVD of VEGF in EA and EC (typeⅡ ) were 0.162±0.061 and 0.179±0.067 (P=0.387), respectively, which were higher than that of NE (P〈0.000). The expression of eIF4E was positively correlated with FIGO stage, differentiation degree and lymph node metastasis in EA, and there was also a positive relationship between the expressions of VEGF with the FIGO stage, differentiation degree, infiltration degree and lymph node metastasis. Positive correlation were observed between eIF4E and VEGF expression of EA (r=0.359, P=0.018). Conclusion: The expressions of eIF4E and VEGF were positively associated with emergence, development and clinicopathologic features of EC, indicating that their expression may be the prognostic factors for EC.
