摘要
目的观察成熟耳石破坏再生过程中,Otoconin 90(OC90)蛋白在内耳的分布表达情况,探讨其在成熟耳石破坏后修复再生过程中的意义。方法采用机械过载作用的方法造成豚鼠耳石破坏模型,免疫组织化学检测耳石破坏后不同时期(0.5~1 h,3 d,7 d)内耳切片各主要部位OC90分布表达情况,并测定椭圆囊囊斑的光密度(OD)值,对其表达强度进行半定量分析。结果 OC90蛋白的分布在耳石破坏后没有显著变化,仍呈现"多点"表达的特点,其在椭圆囊和球囊囊斑感觉上皮的表达于破坏后呈现增强趋势,其中耳石破坏3 d时的OD值增加显著(P﹤0.05)。结论 OC90蛋白在成熟耳石破坏后修复再生过程中起重要作用。
Objective To observe the expression of otoconin 90(OC90) in guinea pigs' inner ear after otoconia damage,and explore its role in otoconia regeneration and repairs.Methods The animal model of otoconia damage was established by mechanical force stimulation,and the slice of inner ear of the guinea pigs in different time(0.5~1 h,3 d,7 d) after otoconia damage were stained by immunohistochemical SP(streptavidin-perosidase) method to detect expression of OC90,the values OD(optical density) in utricle maculae among different groups were measured and compared.Results After mechanical stimulation,expression of OC90 was detected in "multi-site" of the inner ear as in control group,the location of expression showed no changes,but the intensity in utricle maculae increased,value OD of it on 3 d after mechanical stimulation increased significantly(P﹤0.05).Conclusion OC90 plays an important role in otoconium regeneration and repairment.
