6-BA对黑麦草愈伤组织诱导影响的初步研究 被引量:2

摘要 为了确立最佳的黑麦草再生体系,选用多年生黑麦草雅晴、沃土和一年生黑麦草特高等3个不同黑麦草品种的成熟种子为外植体,分别采用CC,MS,NB,N6等4种基本培养基,在每种培养基中分别加入5 mg/L的2,4-D和0,0.05,0.1,0.2和0.4 mg/L等5种浓度的6-BA进行愈伤组织诱导。结果表明:在6-BA浓度为0.05 mg/L时,黑麦草各品种愈伤组织的诱导率最高;在相同浓度6-BA作用下,不同黑麦草品种间愈伤组织诱导率存在显著的品种间差异;不同基本培养基对黑麦草愈伤组织的诱导率也存在影响,4种供试基本培养基中,以CC基本培养基的黑麦草愈伤组织诱导率最高。 The mature seeds of three different ryegrass cultivars, perennial ryegrass Yatsyn and Pandora, and annual ryegrass Tetragold, were selected as explants, four types medium of CC, MS, NB, N6 were used, 5 mg/L of 2, 4- D and different concentration of 6 - BA were added to each medium to study the callus induction in order to establish the regeneration system of ryegrass. The results showed that the callus induction rate of 3 ryegrass cultival~ was highest when the concentration of 6 - BA was O. 05 mg/L. Under the same concentration of 6 - BA, the callus induction rate existed significant differences among different ryegrass varieties. There existed effect of different basic medium on ryegrass callus induction rate, and the ryegrass callus induction rate was highest in CC treatment.
