摘要
为探讨独角莲的体外抗肿瘤作用,将独角莲根茎水提物分别处理人宫颈癌系细胞(HeLa)、人白血病细胞(HL60)、人肺腺癌细胞(SPC)、人骨髓神经母细胞瘤细胞(SH-SY5Y)24、48h后,应用体外细胞培养技术、倒置相差显微镜观察、Cell Counting Kit-8(CCK-8)细胞增殖和毒性实验检测独角莲根茎水提物对各肿瘤细胞的生长抑制作用并筛选出对独角莲根茎水提物最敏感的肿瘤细胞株,运用AV/PI双染流式细胞术对该细胞株生长抑制机理进行初步探讨。结果表明:独角莲根茎水提物对HeLa、HL60、SPC、SH-SY5Y细胞的生长具有抑制作用,与细胞作用48h时的IC50分别为2.575、16.054、2.077、0.862mg.mL-1,与细胞作用24h时的IC50分别为4.303、21.674、3.114、1.158mg.mL-1;倒置相差显微镜观察和流式细胞仪检测结果表明独角莲根茎水提物可能是通过诱导肿瘤细胞发生凋亡来抑制其生长的。
To investigate the anti-cancer effect of the aqueous extract from dried powdered rhizomes of Typhoniura giganteum EngL(AEoTGE) in vitro,several cell lines such as HeLa,HL60,SPC and SH-SY5Y were respectively treated with AEoTGE at 24 and 48h.Then the cellular morphology was observed by phase contrast microscopy,meanwhile the inhibition ratio was measured by CCK-8 assay.The aim was to screen the most sensitive cell lines to AEoTGE among the cancer cell lines.Flow cytometry was used to further investgate the primary mechanisms of cell death.The results showed that AEoTGE inhibited the growth in HeLa,HL60,SPC and SH-SY5Y cells.The IC50 was 2.575mg·mL-1,16.054mg·mL-1,2.077mg·mL-1 and 0.862mg·mL-1,respectively at 48h,while the IC50 was 4.303mg·mL-1,21.674mg·mL-1,3.114mg·mL-1 and 1.158mg·mL-1 at 24h.Moreover,the apoptotic bodies were observed by phase contrast microscopy and the apoptotic cell count were detected by flow cytometry suggesting that AEoTGE could induce apoptosis to inhibit the growth of cancer cells.
