摘要
目的:了解丽水市结核分枝杆菌耐药相关基因rpoB、katG、embB、rpsL和gyrA突变特点。方法:采用PCR-DNA直接测序法,对47株结核菌分离株5个耐药相关基因主要耐药位点突变进行分析。结果:药敏试验共获得耐药结核菌18株,总耐药率为38.30%;序列分析13株存在一个或多个突变位点,未发现碱基的插入或缺失,突变率为27.66%,二者吻合率72.22%。结论:丽水市结核杆菌趋向于耐多种药物,与多个基因突变相关。点突变是本地区结核菌产生耐药性的主要形式。
Objective:To elucidate the characteristics of rpoB, katG, embB, rpsL and gyrA mutation in clinical isolates of Mycobacterium tuberculosis (MTB) from Lishui city. Methods: Mutations of five resistance- related genes in 47 clinical isolates of MTB were analyzed by PCR - DNA. Results: 18 strains of drug - resistant Mycobacterium tuberculosis were obtained in the susceptibility test with the resistance rate of 38.03%. There existed one or more mutations in 13 isolates by sequence analysis. No insertion or deletion mutation was found in these isolates, the mutation rate was 27.66%, the coincidence rate of the two method was 72.22%. Conclusion: The MTB in Lishui tended to be resistant to multiple drugs. Muhidrug -resistance of MTB i- solates was related with the mutation of multiple genes. Point mutation was the main characteristic of drug - resistant MTB in this city.
