摘要
用共培养法 ,以根癌农杆菌质粒为载体 ,携带雄性不育基因和抗卡那霉素基因作报告基因 ,另以该农杆菌携带的抗利福平基因作为农杆菌的筛选培养。以大白菜 ( Brassica chinensis)幼苗的茎尖作为受体 ,转化后的幼苗 ,用卡那霉素 ( 5 0 mg/ L)进行筛选。筛选得到的转化苗经 PCR检测 。
A plasmid vector containing Barnase gene and Km r gene derived from plasmid pBI121 was used for transforming stem tip cells of young seedling of Chinese cabbage.Potential transformed plantlets were selected on MS medium containing 50 mg/L Kanamycin.PCR analysis was used to detect transformed plantlets.
